摘要: | MicroRNA為小片段且無法轉譯的RNA,其片段長度大約為21至24個核苷酸,能夠調控植物與動物體內基因的表現,在植物中能夠影響植物生長發育和抵抗逆境之相關調控。我們利用生物晶片(microarray)的方式去尋找與熱逆境相關的基因,發現AUXIN RESPONSE FACTOR16(ARF16)與AUXIN RESPONSE FACTOR17(ARF17)在熱逆境底下其表現量會降低,此二基因為miR160的目標基因。因此利用Real-time RT-RCR,進一步發現在熱逆境底下,miR160的表現量會上升,並且其目標基因ARF10、ARF16與ARF17的表現量下降。另外也構築了大量表現MIR160a precursor(OE)與降低miR160表現(miR160 target mimicry)的轉殖株。發現在種子萌發率在50oC 2小時熱處理回復四天後,大量表現miR160 OE lines其萌發率都比wild type(WT)來得高,而miR160 target mimicry lines則是比WT來得低。下胚軸在Acquired thermotolerance處理下(38 oC 1.5小時,22 oC 2小時,45 oC 3小時),OE lines之延長率比WT高,Target mimicry lines則反之。另外,在Basal thermotolerance的熱處理下(44 oC 25分鐘),Target mimicry lines之存活率較低,OE lines 較高。簡而言之,我們可以推論阿拉伯芥在熱逆境的情況下,或許能藉由誘導miR160的表現量上升去抑制目標基因(ARF10, ARF16 ,ARF17)的表現,使植物能夠去抵抗熱逆境以求生存。 MicroRNAs (miRNAs) are endogenous riboregulators about 21-24 nucleotides that modulate gene expression in plants and animals. They are non-coding RNAs and play important roles in plant growth, development, abiotic stresses and biotic stresses. However, the involvement of miRNAs in thermotolerance is still unclear. The profile of gene expression was then analyzed by microarray in Arabidopsis under heat stress. Results indicated that the expression of AUXIN RESPONSE FACTOR16 (ARF16) and ARF17 was repressed under heat stress. In addition, miR160’s targets are ARF10, ARF16 and ARF17. Real-time RT-PCR was used to analyze the expression of miR160 and its target genes under heat stress, indicating miR160 was up-regulated and ARF10, ARF16 and ARF17 were down-regulated under heat stress. Transgenic plants over-expressing or target mimicry of miR160 were constructed. The germination and hypocotyl elongation rates of miR160 over-expression and target mimicry lines were better and worse than those of wild type, respectively. Under basal thermotolerance, the survival rates of miR160 over-expression and target mimicry lines were higher and lower than those of wild type, individually. In short, these results showed that miR160 regulating the expression of ARF10, ARF16 and ARF17 might be involved in thermotolerance in Arabidopsis. |
