Tumor-associated macrophages (TAMs) are among the most abundant immune cell populations within the tumor microenvironment (TME) and play critical roles in tumor progression and immune suppression. Our laboratory previously identified zinc and ring finger 1 (ZNRF1), an E3 ubiquitin ligase, as a context-dependent regulator of macrophage inflammatory responses. Although our previous studies using the Lewis lung carcinoma (LLC) model demonstrated that myeloid ZNRF1 regulates TAM polarization and TGF-β signaling, the immunologically “cold” nature of the LLC model limits the evaluation of the role of myeloid ZNRF1 in the crosstalk with adaptive anti-tumor immunity. Therefore, in this study, we investigated the role of macrophage ZNRF1 in the immune-responsive MC38 colorectal tumor model. Clinical analyses suggested that ZNRF1 expression in colorectal tumors is associated with patient survival and T-cell infiltration. Using myeloid-specific ZNRF1-deficient mice, we found that loss of ZNRF1 promoted MC38 tumor growth and reduced infiltration of total T cells and CD8⁺ T cell. Single-cell RNA sequencing further revealed a remodeled TAM landscape characterized by decreased Ccl2⁺Cd74⁺H2-Aa+ inflammatory TAMs and increased C1q⁺Apoe+ mature M2-like TAMs and Mki67+C1q⁺Apoe+ proliferating M2-like TAMs, accompanied by reduced Nkg7+Gzmb+ cytotoxic CD8⁺ T cells and increased regulatory T cells in myeloid-specific ZNRF1-deficient tumors. Mechanistically, ZNRF1-deficient bone marrow-derived macrophages (BMDMs) exhibited enhanced M2-like polarization and reduced surface MHC class II expression, likely through disrupted trafficking of MHC class II-containing secretory lysosome-like vesicles. Overall, this study identifies ZNRF1 as a potential regulator of TAM-mediated anti-tumor immunity in colorectal cancer. |
