Phosphate (Pi) homeostasis in plants is maintained through a coordinated network of sensing, transport, and signaling processes. The Pi transporters PHOSPHATE TRANSPORTER1;1 (PHT1;1) and PHOSPHATE1 (PHO1) play key roles in Pi uptake from soil and xylem loading for root-to-shoot Pi translocation, respectively. Under Pi-sufficient conditions, the ubiquitin E3 ligase NITROGEN LIMITATION ADAPTATION (NLA) maintains Pi balance by mediating the ubiquitination and degradation of PHT1;1, while the ubiquitin-conjugating enzyme PHOSPHATE2 (PHO2) mediates the ubiquitination and degradation of PHO1. Both NLA and PHO1 contain an SPX (SYG1/Pho81/XPR1) domain at their N-termini, which functions as an intracellular Pi sensor by binding inositol phosphates (InsPs), signaling molecules reflecting cellular Pi availability. To examine whether these degradations are InsP-dependent, I generated SPX-domain mutants with disrupted InsP-binding sites. Using a tobacco transient expression system, I demonstrate that NLA-mediated degradation of PHT1 depends on InsP binding, whereas PHO2-mediated degradation of PHO1 occurs independently of SPX-InsP interaction. To further delineate the PHO1-PHO2 regulatory module, truncation assays revealed that PHO2 integrity is essential, as none of its truncation forms could mediate PHO1 degradation. Furthermore, preliminary results from TurboID-based proximity labeling showed that InsP binding may not be required for the interaction between NLA and PHT1. In planta complementation assays revealed that expression of PHO1 variants defective in InsP binding failed to rescue the severe shoot growth and root-to-shoot Pi transport defects of pho1-2. Similarly, expression of the corresponding NLA variants in nla-1 also failed to complement the abnormal shoot Pi levels, demonstrating the crucial role of InsPs in PHO1 and NLA function. This study provides new insights into how InsPs dynamically regulate the protein stability of Pi transporters, which ultimately govern cellular Pi homeostasis in plants.
