摘要: | 本論文主要研究綠色螢光蛋白發光團的衍生物之光物理現象及綠色螢光蛋白發光團衍生物形成凝膠與混摻之凝膠的光物理變化。在本論文的第一章為所研究之題目的基本背景介紹;在第二章中,我們成功地利用取代基電子效應抑制消光之異構化反應的觀念來增強綠色螢光發光團的螢光量子產率,發現間位胺基加鄰位腈基效應可以使螢光量子產率達到 60%;第三章,則是探討綠色螢光蛋白發光團超分子凝膠的環境效應與相關應用性,其中在混摻系統的凝膠可作為光學資訊加密材料的應用,而凝膠分子本身能做為即時細胞螢光顯影材料;第四章為綠色螢光蛋白發光團凝膠結構對異構化反應抑制效應之研究,我們第一個利用超分子自組裝將原生綠色螢光蛋白發光團成功開啟,以及利用溶劑改變超分子自組裝模式調控螢光開啟與關閉。本論文為超分子凝膠與綠色螢光蛋白發光團結合的首例,希望可以開拓一條新的研究方向。 This thesis is about the study of photophysical properties of green fluorescent protein chromophore (GFPc) analogs and its supramolecular organogel systems. The background of this work is provided in Chapter 1. In Chapter 2, the use of electronic substituent effect to slow down the photoisomerization reaction to enhance the fluorescence quantum yield of the unconstrained GFPc is demonstrated, in which a combination of the meta-amino and the ortho-CN substituents results in a quantum efficiency of 60%. In Chapter 3, the effects of gelation on the fluorescence properties of the GFPc either in the liquid phase or in the fibril gel matrix are reported. The applications of the molecular/supramolecular system in optical data encryption/decryption and in real-time cell imaging are demonstrated. Chapter 4 shows the further study of the gel matrix effects on the photo-isomerization reaction of GFPc, in which we observed fluorescence turn-on behavior for the wt-GFP chromophore and solvent-dependent phase transition of fluorescence on/off switching. The results reported in this thesis are pioneer works in GFPc-containing gelating systems and might open up a new research direction. |
