摘要: | 番椒 (Capsicum annuum L.) 為茄科 (Solanaceae) 番椒屬作物。現今商業生產上常是利用細胞質與細胞核雄不稔系統 (Cytoplasmic-Genetic Male Sterility, CMS) 中的雄不稔親 (S/rfrf )、維持親 (N/rfrf ) 與恢復親 (N/RfRf ) 來降低雜交種子的生產成本。為研究稔性恢復基因 (Rf),本研究調查種內雜交之204株F2,利用後裔檢定確認稔性恢復基因之基因型,經適合度檢定符合一對基因的分離比1:2:1。有鑒於SSR分子標記在育種上的應用潛力及其穩定性,近年來開始開發番椒的SSR分子標記。本研究總共篩選337組前人發表的SSR分子標記,與312組透過隨機定序自行探勘的SSR分子標記。使用一組在親本間具多型性之Rf基因連鎖分子標記AFRF3CAPS與112組SSR分子標記建立連鎖群圖譜,將後裔檢定的結果當作一共顯性標記併入分析。本研究建立了15個連鎖群,圖譜總長724.4 cM;比較過去番椒的SSR分子標記圖譜,標記間的連鎖關係與順序普遍吻合。本研究獲得數個與Rf基因連鎖的SSR分子標記,而前人發表所的AFRF3CAPS在本研究中也與Rf基因有連鎖關係;藉由間接對照已公開之番椒遺傳圖譜上的標記,可得知本研究建立的Rf基因連鎖群是位於番椒的第6對染色體上,但目前與Rf基因最近的標記CM458仍距離有21.1 cM。自行隨機探勘的SSR分子標記在圖譜中有群聚的現象,其原因根據前人的細胞遺傳研究,番椒因跳躍子的插入而導致基因體擴增為約番茄的3倍,而番茄的基因體有3/4為異染色質,故隨機探勘的標記若位於基本異染色質會因互換受到抑制而在連鎖群圖譜上產生群聚。未來需繼續開發位於真染色質的分子標記以填補連鎖群圖譜,期望能將連鎖群對應番椒的12對染色體,並進一步研究稔性恢復基因。 In pepper, cytoplasmic-genetic male sterility (CMS) system is very useful for commercial F1 hybrid seed production. Application of molecular markers to monitor the genes involved in CMS system can reduce the time of backcross breeding. Based on the potential utility of SSR markers, the purpose of this research is to construct an SSR-based linkage map of pepper and to map the nuclear Restorer-of-fertility gene. An F2 population consisted of 204 plants was developed as the mapping population. Progeny test was conducted to check the genotype of Rf gene and the phenotype was regarded as a codominant marker. The ratio of segregation of Rf gene is approximated to 1:2:1. The linkage map contains 112 SSR markers and covers 724.4 cM with 15 linkage groups. Compared with the reference genetic map, the linkages and orders of the SSR markers in this study were generally identical to the published maps. AFRF3CAPS, a published Rf-likned marker, is also linked to Rf gene in the study. Our LG6 where Rf gene is located corresponds to the chromosome 6 in a consolidated pepper map indirectly through a published SSR marker, Hpms1-5. However, the genetic distance between Rf gene and the nearest SSR marker, CM458, is 21.1 cM. The future works on this project is to saturate this linkage map by more SSR or SNP markers. |
